In Expository Writing class we were asked to write our first descriptive essay and we were given many topics to choose from. They had to at least have something to do with an experience be it good or bad that had an impact in your life and left a great impression on you. Upon much thought on my part I have decided to write about my first concert that I ever attended and explain why I left an impression on my in three different ways and how it was an enlightening experience for me. Contemporary society deems social class a division within a given population defined by wealth, education, and power, but the lines that divide them unceasingly deepen. Social mobility, or the movement of an individual between the stratification of societal classes, remains virtually illusory, an unattainable falsity that millions have laboriously fought for since the turn of the twentieth century. Monopolies and wage slavery remain definite and palpable, both of which contribute to immobility between social rankings, establishing an unbreakable cycle of poverty. The idyllic ethos of the American Dream, a belief that one will achieve success through hard work and opportunity, prove to be a fallacious, hollow and vague ambition that cannot be attained. Paradigms that exploit the plight of the ââ¬Å"American workerâ⬠beginning in the early nineteen hundreds include muckraker Upton Sinclairââ¬â¢s The Jungle and John Steinbeckââ¬â¢s The Grapes of Wrath. The quandaries that pl ague the characters of the aforementioned novels parallel that of modern day exposà ©s, such as Class Matters, by Bill Keller, Nickel and Dimed, by Barbara Ehrenreich, and Fast Food Nation, by Eric Schlosser, all of which harness and expose the falsehood of The American Dream as a result of wage slavery, class separation, and monopolization of major industries. Domestication Of The Dog Essay Todays dogs serve as a number of different tools. We train dogs to see for the blind, we train them to sniff for drugs, we train them to save peoples lives, and we train them to be our faithful companions. There is no doubt that the dog has a wide variety of skills and jobs. We selectively breed the dog to gain the certain attributes we are seeking, and we know which dogs will perform the best at what we want them to do. The question is how long ago, and why did the dog become our aids, tools, and companions? Answering this question means dealing with the four fields of Anthropology: Ethnologically, Archaeologically, Physically, and Linguistically. The most obvious way to learn about the past of the dog species, is to treat it the same way we treat ancient societies. Archaeologists study where they once were, look at their remains. Where they lived, what they looked like, and how they changed over time. An example of using the Archaeological field of Anthropology would be the excavation of the Roman city, Pompeii, which was destroyed by the volcano Vesuvius in AD 79. When finally excavated, searchers found the remains of a dog lying across a child, apparently trying to protect him. By looking at this individual skeleton, we can estimate that just 1900 years ago, the inhabitants knew of the dogs desire to protect. They probably counted on dogs similarly to the way we do today. Other archaeological digs have suggested that the relationship between dogs and humans dates to about 14,000 years ago. Most experts do agree the dog was the first domesticated animal, was domesticated around 14 to 15,000 years ago. The earliest bones of d ogs that we have recovered come from a site called ein Mallaha in Israel. This site was discovered in 1979 and the bones date back to 12,000 years old but historians believe the dog had been domesticated even a few thousand years before that. Another field of Anthropology is the Ethnological aspect of the science. From looking back at the ancient civilizations customs and beliefs, we can see relationships between their stories and how they lived. This also ties into the linguistic field of Anthropology, where the culture is passed on to the children through stories and myths. An example of this would be the following: historians have studied the Roman myths and legends that were popular in ancient Rome, and they agree that the Ancient Romans relied heavily on watchdogs for protection. The Romans apparently derived this tradition from the legend that a dog guarded the gate to hell. Therefore, they used dogs to protect what was dear to them, homes, valuables, and families. Romans also used dogs for military purposes, some as attack dogs, and some as messengers. They equipped their Mastiffs with light armor and sent them into battle against the enemy, carrying spikes and cauldrons of flaming sulphur (Whitehead 242). These do gs were obviously essential to the ancient Romans lives. The last field of Anthropology that has not been discussed is the physical field. By looking at a culmination of the fossils we have, it not only adds to a holistic approach to the problem, but it also gives us a longitudinal study of a very old question. Instead of gathering the information over multiple visits, which would be impossible, we can get the same information we need by looking at the same object, at different points of history. To do this, we look at fossilized remains that we are able to date. By doing this, we can track the changes we notice. For example, it is quite obvious; that the first domesticated dogs were not as diverse as the dogs we have today. There were only a few kinds of dogs. Fossil remains of the early Bronze Age, 6500 years ago; make it possible to identify 5 major groups of early dogs. As the fossils get younger and younger, we notice a growing of the species. Dogs are obviously mixing and creating new breeds. The wide diversity in breeds that we witn ess today comes from selective breeding as well as natural genetic mutations in the five groups. Physical Anthropology even explores this last point. These natural genetic mutations are causing some dissention in the professional field. A recent study, led by biologist Robert K. Wayne of UCLA, suggests that canines may have been domesticated 100,000 or more years ago, only 30,000 years after the first signs of modern humans. These genetic mutations serve as a harmless ?genetic clock? that indicate the passage of time in the evolution of the wolf to the dog. This same method has been used to show that humans diverged from a common ancestor in North Africa and to show when Asians first entered North America and established Native American populations. Although this idea seems quite rational, most historians will not accept this as fact. The fossils seem to be a much more widely accepted view. People will always accept something they can physically see, rather than theorems and estimat ions. So, as one can see, by using these four fields simultaneously, in a holistic method, Anthropologists can paint a very convincing portrait of a question that seems impossible to answer unless someone was there to witness it. Often, these answers can solve very important questions that provide explanations to why we live the way we do today. Although it is not imperative that we know when and why the wolf became the dog we know today, it is interesting to study the people who helped give us what we know now as mans best companion and protection. If Anthropology can solve this Anthropological problem, it is exciting to think what else we can learn about another group of people or time period, without even being present!Bibliography? There is no secret that the number of manufacturing jobs in the U.S. steel industry has significantly decreased and will probably continue to fall due to several reasons. The United States steel industry entered the period of crises in the late 1950s, which intensively remained until the late 1960s despite the fact that specific measures are taken. Scientific research proves that the decline of the steel industry, which also resulted in the decline of the number of jobs in the steel industry, was caused by the combination of reasons. However, it is also proved the major reason was international trade, or to be exact, the increasing amount of imports. Consequences of the cut down of manufacturing jobs in the steel industry have affected a lot of communities significantly across the country , and especially, Eastern and Midwestern regions of the country, where the majority of steel mills were situated many years ago and continue to be there nowadays. Being a part of deindustrialization process decline of the number of jobs in the U.S. steel industry has negatively affected people, who were laid off their jobs as a result of cut down. Steel mills around which small towns have been built started to close, leaving now opportunities for future well-being for their former employees. Those who did not lose their jobs did not have even the smallest ray of hope and faith that they would not be laid off in the nearest future because the loss of money and the decline of competitive ability of steel mills were evident to everybody. The primary goal of designing current study is to discover and analyze significant reasons, which caused a decline in manufacturing jobs in the United States steel industry. The paper will focus on the historical facts leading up to these reasons as well as consequences of the decrease in the number of jobs. The state of the modern steel industry in the USA will also be described further in the study. 1. Historical Background When the steel industry faced the crisis in 1977-1978, it wasnââ¬â¢t something new and unknown, because the industry had already had problems and hard times starting at the end of the 1950s due to the increase of imports. The state of the steel industry at that time was characterized by prolonged growth, which was even worsened by the rising of imports, lowering profits and constant misunderstandings with the USA government on the matter of pricing policies. By the end of the 1960s, the problem with import became so acute that the leaders in the steel industry had no other choice but to refer to the government, which they tried to persuade to ââ¬Å"negotiate import restrictions with Japanese and European exporters of steelâ⬠[3]. Having acknowledged the necessity to protect domestic producers of steel, the government prepared a so-called ââ¬Å"protectionist legislationâ⬠aiming at eliminating the amount of imported steel into the country. However, taken measures seemed not to be working. Despite the fact that the year of 1974 was rather successful for the steel industry, it was followed by the significant recession in 1975, which ââ¬Å"plunged the industry back into a depressed state ââ¬â a condition from which it has not recoveredâ⬠[3]. What caused the recession? The author of the book ââ¬Å"The U.S. Steel Industry in Recurrent Crisis: Policy Options in a Competitive Worldâ⬠Robert W. Crandall names several reasons, including the ââ¬Å"increase of the environmental costsâ⬠and the ââ¬Å"labor costsâ⬠[3]. As a result of the increase of imports and the failure of the government to control it, a lot of steel mills had either to close down or to decrease the number of workers. The ones that chose to close turned into places like Youngstown, which became ââ¬Å"a place of naked ruins, junk heaps of indiscernible provenance or pint-sized industrial follow-ons, including a few steel processing plants and one ââ¬Å"mini-mi ll,â⬠which doesnââ¬â¢t make steel but simply melts scrap metal into reusable formâ⬠[10]. According to statistical evidence, about 10,000 people were employed there ââ¬Å"before first one; then another big steel company shut down until US Steel struck the final blow in 1979â⬠[10]. In 2002, the mini-mill which still exists and operates in that area employed only ââ¬Å"about 430â⬠workers [10]. Examples of the shutting down of mills and employees dismissal are multiple. At the beginning of the 1980s, the community was shocked when ââ¬Å"U.S. Steel added to the record-breaking chill by announcing it would permanently lay off 15,400 workers and close part or all of twenty-nine mills, the largest shutdown in American historyâ⬠[1]. 2. The first reason is the decision of U.S. government to seize some of the steel mills in the USA at the beginning of the 1950s. This happened on the 8th of April, 1952, when ââ¬Å"President Harry S. Truman announced that, to avert a strike, the federal government was seizing the steel mills of all the major companies involved in a labor dispute with the United Steelworkers of Americaâ⬠[7]. The second reason consists of two interrelated reasons, which have impacted the US steel industry the most and have resulted in the significant cut downs of jobs all over the country. These two reasons include the growth of imports and U.S. governmentââ¬â¢s failure to control it. The steel industry had faced a lot of problems before; however, the real crisis began in 1977. In the middle of 1970s, the industry started its recovery, which was abruptly ââ¬Å"aborted by a sudden surge in imports and the price-cutting associated with this surgeâ⬠[3]. As a result of the increasing amount of imports several major companies closed, the income of the industry lowered to the ââ¬Å"zeroâ⬠mark. It became clear that without governmentââ¬â¢s assistance industry would not be able to survive. The U.S. government understood the importance to design a set of policies ââ¬Å"to ease the pressure on the steel industry and its employees, induce the companies to withdraw or suspend the ir dumping complaints, quiet the congressional proponents of trade protection, and minimize the contribution to domestic inflation, which was rising to 7 percent and beyondâ⬠[3]. However, the plan that was proposed by Anthony Solomon to accomplish all of the goals described above did not address the real needs of the industry, which continued to dismiss its workers by closing down more and more mills. Measures that should have been taken by the government at that period had to be stricter, maybe even in the form of protectionism to help the industry face severe competition from cheaper imports. Instead, the plan that was designed aimed at helping the steel industry ââ¬Å"in its struggle with foreign competitors by assuring ââ¬Å"fairâ⬠competition and reasonable domestic tax and environmental policiesâ⬠[3]. Also, it remains unclear why the U.S. government waited for so long to design these policies, which indeed came out only after nearly 20 years from the beginni ng of the steel industry crisis. And the most awkward consequence of such actions can be seen when the U.S. Army needs the products of the steel industry, and it has to go to the world market to find it. This is how it happened when the U.S. Army found the need for new soldiersââ¬â¢ berets. Instead of buying them from domestic producers, ââ¬Å"it had to go to Communist China to have them made. No U.S. company could produce them to the required quality and specificationsâ⬠[9]. Some of the scientists tried to explain such a weird behavior of the U.S. government. These explanations can be found in the article ââ¬Å"Of fshoring in the Service Sector: Economic Impact and Policy Issuesâ⬠by Alan Garner, who states that ââ¬Å"Economic research finds that protectionism is a costly way to preserve U.S. jobsâ⬠[5]. Instead, the author is convinced that the better way out is to ââ¬Å"adopt policies that ease the reallocation of labor and capital to industries with stronger competitive positionsâ⬠[5]. The sequential reason of lack of government regulations to aid the steel industry, which also contributed to the decline of manufacturing jobs in the U.S. steel industry, is the lack of competitive strength of U.S. mills as compared to Chinese and Japanese mills, for example, which at that time produced more qualitative products and at cheaper prices. Of course, having no or negative profits makes the owners of the steel mills lay off the workers because there is no sense in continuing the operation. The following reason which impacted the number of employees in the U.S. steel industry is deindustrialization. The process of deindustrialization has touched all of the world nations. It is characterized by the reduction of the employment in the manufacturing sector and the decrease of the number of people employed in the service sector of the national economy. Some of the scientists state that the decrease of manufacturing jobs in the steel industry is due to the decrease of the industrial capacity of the steel industry, which is partially true; however, it is necessary not to forget about the lack of competitive ability as compared to imports. Factors which contributed to the process of deindustrialization in the USA include: ââ¬Å"a lack of investment in basic production, plant closings and layoffs, and the large negative merchandise trade balance as evidence that the United States is losing its manufacturing baseâ⬠[6]. People that have been laid off started to shift to the service sector, but those who couldnââ¬â¢t shift had to remain unemployed. The process of deindustrialization does not only mean shifting of the jobs to the service sector. However, it means the decline of the manufacturing industries as a whole, which is followed by the reduction of employment and the decrease of net profits. In conclusion to this point, reasons that contributed to the decline of manufacturing jobs in the United States steel industry include the increase of foreign imports and lack of competitive advantage to resist it; government failure to eliminate the pressure on the U.S. steel industry from the side of foreign producers; deindustrialization and shift of the jobs from industrial sector to service sector of economy; lack of investment in the steel industry; decrease of profits; and bankruptcy of the steel mills. 3. Modern State of the Steel Industry in the USA There is no doubt that the U.S. steel industry will continue to exist and will survive all of the hardships. However, the structure of it is constantly changing, adopting new technologies, changing ownership and adapting to a new competitive environment. It is necessary to acknowledge that ââ¬Å"both changing technology and international competition have contributed to a tougher and more competitive environment for American steelmakingâ⬠[2], but the centers of the steel industry are still situated in the eastern parts of the country. For example, Pennsylvania, Ohio and Indiana employ nearly ââ¬Å"44 percent of all steelworkersâ⬠. In 2004 employment in the steel industry calculated about ââ¬Å"156,000 wage and salary jobsâ⬠[11].The majority of workers are employed by large firms, which can be seen from the following chart. Conclusion Having spoken about the reasons which impacted the number of workers in the U.S. steel industry and contributed to its decline, it is necessary to conclude. As it has been stated above, the process of deindustrialization has been a characteristic feature peculiar to all countries all over the world. And the USA is not an exception at this point. However, not only deindustrialization contributed to the decline of manufacturing jobs in the U.S. steel industry. The U.S. government could have taken more effective measures to protect the steel industry from foreign competition; however, it failed to do it. Besides, the development of modern technologies allowed the industry to reduce the number of jobs by adopting new technologies requiring the minimum of human interference. Also, lack of competitiveness and investment in the sector have made it less profitable leaving the owners of the steel companies no other choice as to reduce the number of steelworkers. Reasons Why Teens Choose to Have an Abortion Teens facing an unplanned pregnancy choose abortion for similar reasons as women in their twenties and thirties. Teens ask the same questions: Do I want this baby? Can I afford to raise a child? How will this impact my life? Am I ready to be a mother? Coming to a Decision A teen considering abortion is influenced by where she lives, her religious beliefs, her relationship with her parents, access to family planning services, and the behavior of her peer group. Her educational level and socioeconomic status also play a role. According to the Guttmacher Institute, the reasons teens most often give for having an abortion are: Not wanting their lives changed by the birth of a babyNot being able to afford a babyNot feeling mature or responsible enough to raise a child Parental Involvement Whether or not a teen opts for abortion often hinges on a parents knowledge and/or participation in the decision-making. Thirty-four states require some form of parental permission or notification for a minor to obtain an abortion. For teens whose parents are unaware that their daughter is sexually active, this is an additional obstacle that makes a difficult decision even more stressful. The majority of teen abortions involve a parent in some way. 60% of minors who have abortions do so with the knowledge of at least one parent, and a large majority of parents support their daughters choice. Continuing Education...or Not The teen who worries that having a baby will change her life has a good reason for concern. Most teen mothers lives are negatively impacted by the birth of a baby; their educational plans are interrupted, which subsequently limits their future earning potential and puts them at greater risk of raising their child in poverty. In comparison, teens who choose abortion are more successful in school and are more likely to graduate and pursue higher education. They typically come from a higher socioeconomic family background than those who give birth and become teen mothers. Even when socioeconomic factors are taken into consideration, pregnant teens are at a huge educational disadvantage. Teen mothers are significantly less likely to complete high school than their peers; only 40% of young women who give birth before age 18 earn a high school diploma as compared to other young women from similar socioeconomic situations who delay childbearing until age 20 or 21. In the long run, the prospects are even grimmer. Less than 2% of teen mothers who give birth before age 18 go on to earn a college degree by the time they turn 30. Access to Abortion Providers Choice is not a choice when theres little or no access to abortion. For many teens in the U.S., obtaining an abortion involves driving out of town and even sometimes out of state. Limited access shuts the door on abortion for those without transportation or resources. According to the Guttmacher Institute, in 2014 90% of counties in the United States had no abortion provider. Estimates of women who obtained abortions in 2005 indicate that 25% traveled at least 50 miles, and 8% traveled more than 100 miles. Eight states were served by fewer than five abortion providers. North Dakota has only one abortion provider. Even when physical access is not an issue, the parental consent/parental notification laws which exist in 34 states in effect limit access for an underage teen unwilling to discuss the decision with a parent. Teen Pregnancy Before Legalized Abortion The fear and hesitancy teens express at the thought of discussing pregnancy with their parents is deeply rooted in our culture. Past generations regarded teen pregnancy as something deeply shameful. Prior to the legalization of abortion, a pregnant girl or young woman was often sent by her family to a home for unwed mothers, a practice that began in the early 20th century and remained until the 1970s. To maintain the secret, friends, and acquaintances were told that the girl in question was staying with a relative. Teens who were afraid to tell their parents they were pregnant often grew desperate to end their pregnancies. Some attempted self-induced abortions with herbs or toxic substances or sharp implements; others sought out illegal back alley abortionists who were rarely medical professionals. Many girls and young women died as a result of these unsafe abortion methods. Lingering Shame With the legalization of abortion with the Roe v. Wade decision in 1972, safe and legal medical means became available to most of the population, and the procedure could be done discreetly and quietly. Although the shame of teen pregnancy lingered, abortion was a way for a teen or young woman to hide her sexual activity and pregnancy from her parents. High school-aged girls who kept their babies were the subject of gossip and pity among students and parents. Media Depictions of Teen Pregnancy and Abortion Today, those views seem strange and outdated to the many teens who choose to become teen mothers. Mainstream media has come a long way in normalizing the idea of teen pregnancy. Films such as Juno and TV series such as The Secret Life of an American Teen feature pregnant teens as the heroines. Much rarer are depictions of teens choosing abortion- a taboo subject in the eyes of Hollywood. Because teen pregnancy has become almost commonplace in many high schools, the pressure to keep it a secret no longer exists as it did in past generations. More and more teens are choosing to give birth, and a type of reverse pressure now exists, with many teens believing that teen motherhood is a desirable situation. The very public pregnancies of famous teens such as Jamie Lynn Spears and Bristol Palin have added to the glamour of teen pregnancy. Thus for some teens, the decision to have an abortion may be a choice that is criticized by peers who only see the excitement of being pregnant and having a baby. Children of Teen Mothers Teens who choose abortion because they recognize their own immaturity and inability to care for a baby are making a responsible decision; it may not be one that everyone agrees with, but it also cuts short a cycle that is on the rise in the U.S. children giving birth to children. More and more studies indicate that children born to teen mothers begin school with significant disadvantages in learning, do poorer in school and on standardized tests, and are much more likely to drop out of school than the children of women whove delayed childbearing until they reach their twenties. Abortion remains a controversial topic, and a pregnant teen considering abortion often finds herself in the proverbial situation of being between a rock and a hard place. But when finances, life circumstances and rocky personal relationships prevent a teen mother from being able to raise her child in a loving, safe, and stable environment, terminating a pregnancy may be her only viable choice. The focus of Huntingtonââ¬â¢s article is the prediction of ideological differences between the different cultures say religions Christianity and Islam being a source of conflict in the world. Summary of the article This article ââ¬Å"Clash of Ignoranceâ⬠by Edward Said aims at faulting the earlier assumption in the ââ¬Å"Clash of civilizationâ⬠where it is only assumed that the major cultures say the West and Islamism are in conflict. The high level of ignorance in the theory and generalization of the West and Islam as cultures is the main thematic concern by Said in this article. The article faults the general ascribed ideology of the West versus the rest as in the cold war period to a now new modern ideology of Islamism versus the West. Critical Discussion Said makes an argument against some of the popular entities in global politics in modern day. The entities are Islamism and the ââ¬Å"Westâ⬠. Said also argues against the idea that Muslims across the globe have an assumption of being a superior culture. This is indeed not a properly sampled and proved theory with empirical evidence of the billions of Muslims across the globe. This can be proved through the embracing of different ââ¬Å"westernâ⬠technologies and the use of the said technologies in the 9/11 attacks on America. Further evidence points at the embracing of western style clothing by some Muslims even the women. The above stated evidence by Said points to the heightened ignorance in factual evidence as regards to calls of existence of conflicts between Islam and the ââ¬Å"Westâ⬠CITATION Sai01 l 1033 (Said, 2001). In this article, Said makes numerous efforts as a result of lack of evidence to show the divide being asserted between the ââ¬Å"Westâ⬠and ââ¬Å"Islamâ⬠. He points out the different societies across the world where both sides in this alleged divide live in harmony. The increased level of anti-Islamic perspectives s portrayed in the media in the ââ¬Å"Westâ⬠is enough evidence of the level of hate speech directed towards Muslims who stay in the ââ¬Å"Westâ⬠. Said contents that the genesis of such anti Islamism begun in Europe in the wake of the destruction of Roman Empire by the Arab conquest. This led to the destruction of Christian unity at that time and now the resentment against Muslims has now been reignited. Positive aspects of the past relationship between the two cultures have been overshadowed by this loathe towards Islam. For instance the west embraced science, sociology and philosophy of Islam yet now they seen to cast negative aspersions about the religion in general based on a few activities of terrorists in the name of religion CITATION Sai01 l 1033 (Said, 2001). In most parts of the article, Said revisits the similarities between the two sides of the alleged divide and accuses the West of having selective memory and double standards. This is based on some of the atrocities committed by the West that are similar to the September 11 attacks in the name of religion. Examples of such atrocities include the Branch Davidians and the supporters of Jim Jones the reverend. Said further attacks the use of powerful idioms in print media in the ââ¬Å"Westâ⬠since most of these articles do not appreciate the complicated histories between the two sides. His demonstration of the lack of distinction between the two sides and the successful integration of the two cultures in clothing, adoption of technology and co-existence in some societies show the lack of conflict between the two cultures. However, his subjective statements across the article taint his argument. His critic of Huntingtonââ¬â¢s book ââ¬Å"The Root of Muslim Rageâ⬠was personal as he says Huntington did confuse himself by showing he is such a clumsy thinker. Generally, this article ââ¬Å"The Clash of Ignoranceâ⬠is quite a persuasive piece of work and convincing. Conclusion From the analysis explored in this article, the different stakeholders must unite to fight the increasing divide between the two sides of the divide. How does Betchman portray the weakness of English Middle Classes in Westminster Abbey? In the poem 'In Westminster Abbey' the poet (John Betchman) speaks in from the view of a lady living in the English middle classes in the period, in which the Second World War took place. The poet has set the poem in the head of the heart of England's church, Westminster Abbey. The poet narrates to the reads the feelings of this lady and her feelings of the war, but what the poet does in mainly stereotype all women in the English middle classes at the time. In the following I shall talk about how poet portrays the weakness of the English middle classes. The poem starts of with the lady talking to god and continues through out with this lady talking to god. The poet sustains a continues rhythm through out with a regular four beats in each line, for example 'LET me TAKE this OTHER glove OFF' , this technique retains the readers interest in the poem since the this technique has been used consistently through out the poem. An additional technique that the poet has used is couplets this is another method that stays persistent through the poem, for example in the first stanza lines two and four; 'Let me take this other glove off As the vox Humana swells And the beauteous fields of Eden Bask beneath the Abbey bells.' This is another example of the poet trying to keep the reader attracted to the poem through out, and keep a persistent flow. As one reads the poem they expected to realize that that the lady has a selfish attitude, this is shown in many aspects of the poem one of which is shown in the second stanza; 'Gracious Lord, OH BOMB THE GERMANS, Spare their women for thy sake, If that is not too ea... ...mpire, this I think the poet also narrates this fact in the third stanza; 'Keep our Empire undismembered Guide our forces by Thy Hand,' Here the lady is asking God keep the Empire united and to guide them to victory. In conclusion the poet portrays a major weakness of the English middle classes in the poem 'In Westminster Abbey', although they were wealthy, well educated for the time's standards and considered as of a superior status than others. One is led to believe that they lacked the understanding and wisdom that people generally have today, also they were selfish and ironic which have learnt from lines like 'Don't let anyone bomb me' and 'If that is not too easy'. I have made these statements from the assumption that the reader is led to believe that this poem is a stereotype and a generalized view or opinion of the English middle classes.

New Horizons classes will resume on a regular schedule during the Hampton City Schools exam week. Students will therefore be excused from New Horizons classes when they have exams at Hampton High School, but the student needs to share that with the New Horizons teacher in advance. Students may attend New Horizons when they do not have exams at Hampton High School. Please avoid scheduling dental and medical appointments during the exam period.If illness or some other emergency arises, which prevents a student from being in attendance for a scheduled exam, parents/guardians must communicate with the appropriate teacher(s) to reschedule. If justification for missing an exam cannot be validated and/or no communication occurs between the parent and the school, the student shall be considered unexcused and will receive a ââ¬Å"Oâ⬠for the semester exam, regardless of the class average. The Attendance Office may be reached at 896-5882 for emergencies during the exam period.If a studen t arrives with an unexcused tardy to an exam period, he/she may have the remaining time to complete the exam, however additional time will not be provided. If a student arrives with an excused tardy, the teacher may offer additional time prior to the completion of the exam week so that the student has the full 2 hours as needed. Transportation will be the student's responsibility. Monday, Jan aura 26th, and Tuesday, January 27th, will be Teacher Workdays. Students will not report on either day.The second semester will begin on a RED Day on January 28th. All 1st block class locations will be posted on the walls. The classical theory or classicism emphasises the notion of individual rights, the importance of free will and the rule of law (White and Haines, 1996). It also suggests humans are essentially self-seeking and self interested, and hence will make their decisions simply by choice. Criminologists such as Cesare Beccaria and Jeremy Bentham were contributors to the classical theory (Maguire, Morgan, Reiner, 1997). The biological theories are loosely based on the scientific study of criminals and criminal behavior. The most prominent and influential biological criminologist was Cesare Lombrosso who argued that criminals are essentially born, not made. He based his concept heavily on the theories of evolution as he attempted to distinguish different types of individuals on the basis of racial and biological differences (Maguire, Morgan, Reiner, 1997). Closely linked with this theory is that of psychological positivism. Psychological positivism directs its argument to the mind of the crimin al and thus we encounter the notion of the ââ¬Ëcriminal mindââ¬â¢ (Burke, 2001). Within this theory there are three broad categories of psychological theories of crime. The first two categories are concerned with psychodynamic and behavioral learning theories, the third associated with cognitive learning theories. The likes of Sigmund Freud, Ivan P Pavlov, B.F. Skinner, Edward Tolman and Jean Piaget were all influential and prominent psychological criminologists of their time (Burke, 2001). The last theory, sociological positivism is arguably the most logical theory of the four, recognizing the social factors external to the human being, placing constraints on the given personââ¬â¢s choice of action. Circumstance and social pressure is taken into consideration when examining crime rather than placing reason on other factors such as biological/psychological positivism (Gibbons 1979). Prominent sociological criminologists include the work Emile Durkheim. This essay will discu ss the above criminological theories and determine which theory provides the best explanation to crime. The school of classicism or classical theory was introduced in the eighteenth century providing the first naturalistic explanation of crime (Moyer, 2001). The cruel punishments in medieval Europe were superseded by the newly implemented classical theory, which was heavily influenced by the eighteenth-century intellectual movement (Gibbons, 1979). The classical theory was the dominant perspective for around a century however; later fell into disrepute, particularly due to the new wave of American criminologists and positivism. Classical theory is however, still highly regarded, as it represented a remarkable humanitarian reform and provided fundamental rationale for many criminal codes of the western world (Gottfredson and Hirschi, 1990). The Criminologist Cesare Beccaria is seen today as the father of classical criminology. Beccariaââ¬â¢s career began when he joined with two friends to form a club in which discussed topics of literary and social interest. Beccaria published a crit ical essay on administrative law, known as Dei deliti e delle pene (On Crimes and Punishments). It became an immediate success, however Beccaria initially remained anonymous, as the cost of criticizing the church and state could be harsh and brutal (Gottfredson and Hirschi, 1990). His essay put forward the central principals of the classical school of criminology, which were ââ¬Å"practically all of the important reforms in the administration of criminal justiceâ⬠(Beccaria, 1767). Another well-known contributor to the theory of classicism was Jeremy Bentham, who was seen as a modern version of Beccaria (White, Haines, 1996). Bentham argued than humans are rational beings and thus implied that as humans, we all have free will. He believed that lawmaking should aim to achieve happiness for the majority of people under his idea of ââ¬Ëutilitarianismââ¬â¢ (Moyer, 2001). The classical school represents the emergence of modern criminological thinking, dismissing earlier theor ies that crime was a supernatural phenomenon (Moyer, 2001). Classicism provides somewhat of a clear explanation to crime however, other factors should be considered when examining crime rather than simply individualistic actions, as sometimes we make choice because we have to- not because we want to. Classicism was soon out shadowed by the emergence of positivism, as science was introduced to the criminological explanations of crime. Biological positivism is based on the scientific understanding of crime and criminality and how factors and forces beyond the immediate control of the individual shape it. It assumes a biological distinction between the ââ¬Ënormalââ¬â¢ and the ââ¬Ëdeviantââ¬â¢ (White and Haines, 1996). Cesare Lombroso who is generally acknowledged as the founder of positive criminology first popularized it, as he claimed that criminal and deviant individuals are the way they are due to their biological composition (Burke, 2001). Lombroso adopting a social Darwinian perspective, suggested that humans can be grouped at various levels of development (Moyer, 2001). He contended that certain characteristics were indicators of an atavistic person. An atavistic person is said to possess physical, psychic, or functional qualities of remote and more primitive ance stors (Moyer, 2001). He classified these individuals as ââ¬Ëborn criminalsââ¬â¢ and hence came up with the theory; ââ¬Ëthe criminal is born, not madeââ¬â¢ (white, Haines, 1996). Lombroso then divided criminals into four broad categories. The first category, born criminals, was distinguished by their physical atavistic characteristics (Burke, 2001). The second and third categories included insane criminals as well as criminals or criminaloids. This included those who commit crimes in response to available opportunities. However, these categories also encompass traits that predispose them to criminal behavior (Burke, 2001). The final category, criminals of passion, is based on the premise that crime is motivated by anger, love or honor. Many criminologists today however, consider Lombrosoââ¬â¢s approach to crime to be simplistic and naà ¯ve. This theory became unpalatable for many in the context of the mid-twentieth century instances of mass systematic extermination of certain groups based on, for example, ethnicity, sexuality or their health (Burke, 2001). Subsequently biological positivism does not provide a plausible or logical explanation of crime. Linked closely to biological positivism is psychological positivism. Psychological positivism is based on the understanding that the mind is responsible for acts of crime and criminality and thus we encounter the notion of the ââ¬Ëcriminal mindââ¬â¢ (Burke, 2001). However, as mentioned, there are three broad categories of psychological theories of crime. The first category: the psychodynamic theory; was popularized by the extremely influential work of Sigmund Freud. His assertion that ââ¬Å"sexuality is present from birth and has a subsequent course of development is the fundamental basis of psychoanalysisâ⬠and one that has provoked much controversy (Burke, 2001). Freud proposed that personality is comprised of three separate components. The first section is the id, which are the primitive biological drives that underlie behavior (Maguire, Morgan, Reiner, 1997). The id is the ââ¬Å"unconscious aspect of personality that leads the individual to seek self-gratificationâ⠬ (Brown, Esbensne, Geis, 1991). Secondly, the superego or ââ¬Ëthe consciousââ¬â¢, which operates in the unconscious, yet consists of values that are internalized through oneââ¬â¢s early interactions, usually with their parents. The superego represents the moral and ethical standards of society (Gottfredson and Hirschi, 1990). The third is the ego or the conscious personality. The ego is the mediating force that arbitrates between the pulls of the id and the superego. The ââ¬Ënormalââ¬â¢ person was said to comprise these three components representing a ââ¬Å"balanced conflictâ⬠(Burke, 2001). The second category: behavioral learning theories; originate from the works of criminologists Ivan. P. Pavlov and B.F. Skinner. Pavlov famously studied the process involved in simple, automatic animal behaviors, in particular salivation in the presence of food (Burke, 2001). Pavlov carried out a series of tests and found that responses that occur spontaneously to the nat ural (or unconditioned) stimulus, could be made to happen (conditioned) to a stimulus that was previously neutral. He focused on the process of turning on a light just before feeding the animal. Eventually the animal would salivate when the light comes on regardless of food being there or not. This process is called extinction (Gottfredson and Hirschi, 1990). While Pavlovââ¬â¢s work concerned automatic behaviors occurring in response to stimuli, B.F Skinner extended the same principal to active learning. This involved the animal having to do something in order to obtain a reward or avoid a punishment (Burke, 2001). They found that learned behaviors are far more resistant to extinction if the reinforcement has only been used occasionally throughout learning (Burke, 2001). This theory makes sense, for example, if you put money into a ticket machine but donââ¬â¢t receive a ticket, you simply stop using the machine, whereas many people put money into gaming machines despite their slight chance of obtaining a prize (Burke, 2001). The third category: cognitive learning theory; has itââ¬â¢s foundations in a fundamental critique of the predestined actor model. Edward Tolman and Jean Piaget proposed, ââ¬Å"by observing the responses that individuals make to different stimulus conditions, it is possible to draw inferences about the nature of the internal cognitive processes that produce those responsesâ⬠(Brown, Esbensne, Geis, 1991). These psychologists emphasized the importance of the organizational process in perception, problem-solving and learning. From this they proposed that individuals were predisposed to organize information in particular ways (Maguire, Morgan, Reiner, 1997). However in recent years it has been acknowledged that criminals have some degree of choice. They can choose to imitate the behaviors of others or choose not to. There also may be a number of variable factors to influence this choice and hence, it can be concluded that crime is not inevitably destiny. Consequently, psychological positivism does not provide an adequate explanation of crime (Burke, 2001). The Sociological school has been extremely influential and is considered by many prominent criminologists today as the most logical, common sense, and, if partial explanation of crime and criminal behavior (Kohn, 2004). The sociological school essentially rejects the above individualist explanations of crime. It recognizes that crime is a socially constructed occurrence, however ââ¬Å"acknowledges the threat it places on the continuance of the given society and thus needs to be controlledâ⬠(Freund, 1969). Emile Durkheim was known as the founding father of academic sociology in France and consequently extremely influential with sociological theories. Durkheim presented two central arguments to explain the growth of crime and criminal behavior (Burke, 2001). Firstly, he suggested that such societies encourage a state of unbridled ââ¬Ëegoismââ¬â¢ that is different to ââ¬Å"the maintenance of social solidarity and conformity to the lawâ⬠(Burke, 2001). This describes p re-industrial types of society where individuals encompass similar or the same skills, beliefs, customs, religion or even work tasks (White, Haines, 1996). From this, certain patterns or trends could be acknowledged and noted. For example, Durkheimââ¬â¢s famous study of suicide demonstrated empirically that suicide rates vary depending whether or not the country is predominantly Catholic or Protestant. The point he made was that suicide could not be explained based on individualist actions or psychological factors, as it is a ââ¬Ësocialââ¬â¢ phenomenon (White, Haines, 1996). The second claim suggested the likelihood of inefficient regulation is greater at a time of rapid modernization. This is because new forms of control have not evolved sufficiently to replace the older, and now less appropriate, means of maintaining solidarity (Burke, 2001). However, Durkheimââ¬â¢s predecessor, Auguste Comte, is responsible for much of the work interpreted as his own. The two shared very similar views, however, the essential difference between the two was their differing views on human nature. Comte believed the human being has a natural and inherent desire to reach perfection whereas Durkheim rejects this view and proposes a ââ¬Ëdualisticââ¬â¢ view of human nature. He proposed a duality between the needs of the body and the soul (White, Haines, 1996). The sociological school highlights aspects of offending which were clearly absent from psychological and classical theories. It acknowledges that the relationship between lack of opportunity, alienation and criminal behavior is more important in the current economic climate, as youth unemployment is high and the inequalities and divisions between the rich and the poor are continuing to grow (White, Haines, 1996). Rather than simply placing the entire burden on the criminal or their individualistic actions, the sociological school considers other factors, providing a coherent and logical explanation of crim e. The four schools of thought all provide differing explanations to crime. Classicism is based on the premise of free will and places entire responsibility on the criminal. Biological positivism differs in the sense that it places the responsibility on forces beyond control of the criminal and hence, suggests that the criminal is born, not made. Similarly psychological positivism puts emphasis on the mind as their explanation of crime as we encounter the ââ¬Ëcriminal mindââ¬â¢. However, the sociological school rejects individualist explanations of crime and considers social factors surrounding the criminal providing the most common sense explanation of crime. After examining the above theories it can be concluded that classicism, biological and psychological positivism are all heavily flawed to varying degrees and it is very difficult to establish any consistency within them. It Took a While, but the Midget Raisin is No More Responding to a petition first filed by the Little People of America in May 2103, the U.S. Department of Agriculture has agreed to remove ââ¬Å"midgetâ⬠from its list of size classifications for raisins.à In a new federal regulation proposed by the USDAââ¬â¢s Agricultural Marketing Service (AMS) on August 13, the USDA would eliminate all five occurrences of the word ââ¬Å"midgetâ⬠from the ââ¬Å"U.S. Standards for Grades of Processed Raisins.â⬠The USDA had used ââ¬Å"midgetâ⬠to describe the smallest size of commercially processed raisins since WWII. ââ¬ËSmall,ââ¬â¢ Not ââ¬ËMidgetââ¬â¢ ââ¬Å"The action would clarify AMS grade standards by eliminating the use of the term ââ¬Ëmidget,ââ¬â¢ while consistently using the term ââ¬Ësmallââ¬â¢ for raisins graded in that category,â⬠states the USDAââ¬â¢s proposed rule. ââ¬Å"The industry has used the two grade terms interchangeably for years. The proposed grade standards would be applied uniformly by all handlers.â⬠The USDA acknowledged that the change was being made in response to the petition from the Little People of America (LPA), a non-profit advocacy group providing support and information to persons with a medical diagnosis of dwarfism or other form of short stature between 2ââ¬â¢-8â⬠and 4ââ¬â¢-8â⬠tall and their families. ââ¬Å"On May 13, 2013, AMS received a petition from the Little People of America stating that they ââ¬Ëare trying to raise awareness around and eliminate the use of the word midget,ââ¬â¢Ã¢â¬ the USDA said. According to the USDA, the petition also stated that, ââ¬Å"Though the use of the word midget by the USDA when classifying certain food products is benign, Little People of America, and the dwarfism community, hopes that the USDA would consider phasing out the term midget.â⬠The Problem with ââ¬ËMidgetââ¬â¢ The LPA considers the word ââ¬Å"midgetâ⬠to be ââ¬Å"an antiquated slang term often used as slur toward shorter than average persons, specifically, ââ¬Å"a person with a diagnosable skeletal dysplasia or medical condition,â⬠according to the groupââ¬â¢s Web site. In 2014, the LPA criticized Marvin Lewis, head coach of pro footballââ¬â¢s Cincinnati Bengals for referring to Cleveland Browns quarterback Johnny Manziel as a ââ¬Å"midget.â⬠Manziel, while considered by some in the sport to be ââ¬Å"shortâ⬠for a pro football quarterback, is 6ââ¬â¢-0â⬠tall. ââ¬Å"LPA has been actively working to get the word taken out of societys vernacular, where it is often used carelessly and without regard to who else it may affect,â⬠states the LPA. Dwarfism is a recognized condition under the Americans with Disabilities Act (ADA). In 2011, Startbucks Coffee agreed to pay $75,000 to settle an ADA-based disability discrimination lawsuit brought by the U.S. Equal Employment Opportunity Commission (EEOC) charging that the coffee retailing giant had unlawfully denied a reasonable accommodation to a barista with dwarfism at one of its El Paso stores and subsequently fired her because of her disability. How Tall is a Midget Raisin? Under USDA standards adopted in 1978, midget ââ¬â now to be called ââ¬Å"smallâ⬠raisins are ââ¬Å"95 percent, by weight, of all the raisins will pass through round perforations 24/64-inch in diameter, and not less than 70 percent, by weight, of all raisins will pass through round perforations 22/64-inch in diameter.â⬠Size standards for commercially-processed raisins are set by the USDAââ¬â¢s Raisin Administrative Committee, which had already ââ¬Å"approved the removal of the term midget from the standardsâ⬠in 2014. When Will You Notice the Change? While you might already see ââ¬Å"smallâ⬠replacing ââ¬Å"midgetâ⬠on raisin packaging and advertising, the change will not become official for while. As required by law, the USDA must continue to accept public comments on the new regulation until October 20. At least a month later, the new regulation will be published in the Federal Register, making the change from ââ¬Å"midgetâ⬠to ââ¬Å"smallâ⬠official. I have been performing a double mutant screen using an RNAi knockout library. Cep-1 is a protein that causes apoptosis due to DNA damage in the worm. The cep-1 protein is an ortholog of the human tumor suppressor protein p53, which is found mutated in many cancerous cells. The aim of my experiment is to find genes that are regulators of cep-1 which cause lethality if mutated with cep-1. Alternatively mutants which are lethal as single mutants but survive as double mutants in combination with a cep-1 mutant are also potential genes of interest. These mutants have the potential to be regulated by drugs/proteins to change the levels of cep-1 and induce/prevent apoptosis in cells. Under normal conditions a cell has regulatory proteins and check points that make sure that a cell is growing at the right pace and if for any reason the cell starts to grow irregularly then these proteins stop the cell growth and cause apoptosis. Apoptosis is the programmed death of a cell, as you can imagine it is tightly regulated and loss of regulation can result in catastrophe for the cell and the organism. When apoptosis is over active, healthy cells will die and this is often seen in diseases such as neurodegenerative diseases, hematologic diseases and many other tissue damage diseases. Loss of apoptosis in turn results in cancers, autoimmune diseases and inflammatory diseases. P53 is a tumor suppressor protein that was discovered in 1979. Since then a lot of research has been done into p53 and its functions. P53 is a tumor suppressor protein that is involved in defense of the cell. It is activated when a cell undergoes many various kinds of stress, such as DNA damage, hypoxia, metabolic stress and oncogene activation. It is one of the most important barriers to cancer in many mammals. P53 works in many various pathways, primarily p53 is involved in binding to transcription factors that then activate pathways involved in cellular defense, such as preventing angiogenesis and cellular growth. Malfunctioning p53 is one of the greatest hallmarks of cancer. The majority of mutations found in p53 are due to single nucleotide substitutions in the amino acids of the DNA binding domain of the protein. Similar cancerous phenotypes are observed when either p53 loses function due to a loss of function mutation or when negative regulators of p53 are overactive or positive regulators of p53 lose their function. However p53 is quite unique amongst tumor suppressor proteins because different missense mutations in the protein can cause different levels of p53 activity and hence have varying effects on the host. The effect of the mutation is further modified depending on the genetic background of the person with the mutation. Due to the different effects of various amino acid substitutions and the varying genetic background of patients it is quite difficult to study large populations. This is because high-throughput sequencing and genome wide single nucleotide polymorphism maps are expensive to obtain. The prices are going down as new technologies are becoming available however until now there have been no larger scale studies relating different mutations to varying levels of risks for different types of cancers. It is also hard to perform molecular studies on entire humans and so research is done on cell lines, mice and on cep-1 the Caenorhabditis elegans ortholog of p53. Caenorhabditis elegans is a worm in the nematodes phylum. They are a very well-studied organism due to their ease of study. C. Elegans are one of the simplest organisms that have a nervous system and that make them a very good model organism for neuronal studies. They are transparent creatures and so many molecular and cellular processes especially those involved in development have been thoroughly researched. They have a short generation time and are very cheap to maintain. They are an extremely good model organism for performing screens because any of their nonessential genes can be knocked out easily by using RNAi. The cep-1/p53 pathway is highly conserved throughout evolution as it is an extremely important pathway that is essential for cellular survival over time. The cep-1 protein works through the following pathway to cause apoptosis in cells suffering DNA damage. Cep-1 à ¯Ãâà Egl-1/Ced-13 ââ¬â| Ced-9 ââ¬â| Ced-4 à ¯Ãâà Ced-3 à ¯Ãâà Apoptosis A similar pathway is observed in humans p53 à ¯Ãâà BH3 ââ¬â| Bcl2 ââ¬â| Apaf1 à ¯Ãâà Caspase à ¯Ãâà Apoptosis A majority of these proteins are orthologs to one another. It was hoped that through my screen further proteins which interact with Cep-1 would be found and then their orthologs in humans could be found and then researched and targeted to regulate p53. Materials and Methods The experiment in itself was an extremely simple but time consuming experiment. Screens were performed with mutant worm strains which were then fed RNAi, through Escherichia coli (E. coli), to silence the gene of interest and create double mutants. To begin with a liquid screen is preferred over a solid screen. This is because with a liquid screen you can screen a larger sample of double mutants more easily than a solid screen. Liquid Screen The following strains of bacteria and worms were utilized, E. Coli ââ¬â OP50, C. Elegans ââ¬â N2, C. Elegans ââ¬â GK138, C. Elegans ââ¬â LG12501. E. Coli ââ¬â OP50: Food source for C. Elegans C. Elegans ââ¬â N2: Wild type worms C. Elegans ââ¬â GK138: Cep-1 Mutant worms C. Elegans ââ¬â LG12501: Cep-1 Mutant worms RNAi Knockout library for C. Elegans Chromosome 1 Day 1: Grow worms on Nematode Growth Medium (NGM) plates with OP50 as a food source for the worms. Day 3: Bleach worms that were plated on day 1 so as to only have eggs remaining on plates. Bend glass rod into L shape using a high temperature flame (Bunsen burner) Pipette bleach onto plates with worms on them and gently scrape the surface to mix worms and eggs with bleach Pipette fluid into Eppendorf tube and centrifuge at max speed for 3 minutes Aspirate most of the fluid and keep as much of the pellet as possible Suspend pellet with bleach Repeat steps 3 and 4 Suspend pellet using M9 solution Centrifuge at high speed for 1 minute Aspirate most of the fluid and keep as much of the pellet as possible Repeat 7 and 8 Suspend pellet using M9 solution and vortex Eppendorf tube at low speed to mix solution Place Eppendorf tubes in slow rocker in a 20à ° fridge overnight to allow eggs to hatch Day 4: Replicate bacteria containing RNAi from knockout library using a sterile 96 pin replication tool into a 96 well containing Liquid Broth (LB) with Ampicillin and allow the bacteria to replicate overnight at 37à °C in an incubator Induce transcription of RNAi using adding 0.1 Molar IPTG into wells and place in a shaker for 1 hour. Pellet bacteria by centrifuging in a cold centrifuge at 5à °C for 5 minutes at 2,500 g Remove the supernatant by flipping over the well quickly but carefully so as to keep pelleted bacteria in the wells Suspend bacterial pellet in wells using NGM Pipette worms into wells and place in 37à °C shaker Day 8: Remove worms from shaker gently making sure not to tilt the 96 well plates Observe and record phenotype of the worms Compare phenotype between the 3 different strains of worms Leave worms in 20à °C fridge overnight Day 9: Remove worms from fridge gently making sure not to tilt the 96 well plates Observe and record phenotype of the worms Compare phenotype from previous day Compare phenotype between the 3 different strains of worms Sequencing After potential hits were found in the liquid screen the RNAi from these bacteria were sequenced to ensure that the sequence of the RNAi was correct and hadnââ¬â¢t randomly mutated over time. RNAi was prepared by using a Qiagen miniprep spin kit. Suspend bacteria in 250 à µl Buffer P1 and place in a microcentrifuge tube Mix 250 à µl Buffer P2 and shake the mixture by flipping tube over a few times To the mixture add 350 à µl N3 buffer, mix well quickly Centrifuge at 13000 rpm for ten minutes Aspirate supernatant into new tube Centrifuge again for a roughly 1 minute and discard the flow through Wash spin column with 0.5 ml PB buffer and centrifuge for 1 minute, discard flow through Wash spin column with 0.75 ml PE buffer and centrifuge for 1 minute Discard flow through and centrifuge at maximum speed for 1 minute Place prep column in a sterilized 1.5 ml microcentrifuge tube Add 50 à ¼l of water to prep spin column and allow to rest for 1 minute after which centrifuge for 1 minute After the Qiagen miniprep is complete the tubes were sent for sequencing to The Centre for Applied Genomics where it was sequenced and results were obtained within a week. Solid Screen Solid screens were performed on genes which were found to have increased lethality with cep-1 deletion or increased survivability with cep-1 deletion. The following strains of bacteria and worms were utilized, E. coli ââ¬â OP50, C. Elegans ââ¬â N2, C. Elegans ââ¬â TG12 E. Coli ââ¬â OP50: Food source for C. Elegans C. Elegans ââ¬â N2: Wild type worms C. Elegans ââ¬â TG12: Cep-1 Fluorescent tagged (GFP) worms C. Elegans ââ¬â Ned-8: Positive control C. Elegans ââ¬â HT115: Negative control RNAi Knockout library for C. Elegans Chromosome 1 Day 1) Streak RNAi bacteria of interest from RNAi Library to obtain single colonies. Allow them to grow overnight in 37à °C incubator Day 2) Pick and grow a single colony in 5 ml of LB + Amp + Tet overnight in a 37à °C shaker Day 3) Add 0.1M IPTG for 4 hours to induce RNAi Plate 100 à µl of induced bacteria on RNAi plates incubate at 37à °C incubator overnight Day 4) Pick 5 worms at the same stage and plate onto RNAi plates Allow to grow over 4 days at 20à °C Day 8) Score phenotypes and compared between different strains Day 9) Score phenotypes again on next day and compare between different strains and the previous day Fluorescent Microscopy We used a Differential interference contrast (DIC) microscopy to observe localization of cep-1 in TG12 worms. Worms were mounted using the following method Place a drop or two of hot liquid agarose onto microscope slide Immediately place a second slide perpendicularly on top of the agarose Allow agarose to settle for one to three minutes Gently slide off second slide so as to leave an agarose patch behind Add a drop of 1mM Levamisole to paralyze worms and prevent their movement Pick worms and place them on the slide Slowly place coverslip on top of worms, be very gentle Once prepared the slides were observed using a DIC microscope to find any irregularities in the localization or amount of cep-1 in the worms, especially in the germline and the eggs. Results The aim of this experiment was to find genes that cause lethality as double mutants with cep-1 mutants but not in wild type worms, these genes would be positive regulators of cep-1 and cause apoptosis through cep-1. The screen was also designed to find worms that cause lethality in wild type worms but not in cep-1 mutants, these would be negative regulators of cep-1 and cause apoptosis through cep-1. The results were gathered and tabulated to allow for an easier and better analysis of data. Note: There is a lot of data and so only data that is relevant is shown. These 3 hits were then repeated multiple times on solid media to make sure the results were accurate.
